In a deployment scenario, client computers are behind a firewall. In the case that the firewall is performing NAT for the clients, what happens to the Persistence algorithm (under Outbound Policy) with Persistence mode set to “By Source”? Would that imply that all clients will go to the same ISP as they share the IP after NAT?

You are correct and for this scenario persistence “by destination” may allow load balancing to occur using the persistence algorithm.
