and speed fusion layer 3 same time works on same peplink 580/380 ?
Yes. So lomg as the L2 VPN is to a different VLAN than the L3 VPN. Use internal firewall rules section to do this.

can i do this in the edge? i trasparent 3 vlan in trunk can i made firewall rule to each vlan ?
